What is the significance of the water that gushed from Jesus' side when his side was pierced?

The significance is that it forensically demonstrated that Jesus was dead.

Right. When a person dies, the red blood cells separate out from the watery plasma. The people back then might not have known that but it's proof we understand today that death has occurred.

Actually, there is ample proof to show that they did know that that water mixed with blood was an evidence of death - even though they had no understanding of the human cell.

the significance might have been that the ark of the covenant was hidden in Jeremiah's grotto, which were some hidden caves directly below Golgotha.
and Christ was stabbed, and out of His side (JN 19:34) came forth blood and water.
and then MT 27:51 the rocks were torn rent.

the blood was sprinkled on the mercy seat.
